The Sequence Card Game accommodates 2 to 8 players, who are divided into two, three, or four teams with equal numbers of participants. Each team is assigned a distinct color of chips, with a maximum of four players per team and up to four teams in total.

The game board features each card illustrated twice; however, Jacks are not depicted on the board, although they hold strategic significance during gameplay.

During their turn, a player selects a card from their hand and places a corresponding chip on the matching space on the game board. For example, if a player chooses the Ace of Diamonds from their hand, they place a chip on the Ace of Diamonds space. Jacks possess special abilities: Two-Eyed Jacks act as wild cards able to represent any card and can be used to occupy any open space on the board. One-Eyed Jacks can remove an opponent’s token from a space, enabling players to block opponents or disrupt ongoing sequences. It is important to note that neither type of Jack can be used to remove a chip that is part of a completed sequence; once a sequence has been successfully created by a player or team, it remains unaltered.

Following the completion of their move, the player draws a new card from the deck. Players may place chips on any of the matching card spaces that are not already occupied by an opponent’s marker. If a player holds a card with no available spaces on the board, the card becomes "dead" and can be exchanged for a new one during their turn. To do so, the player discards the dead card by placing it in the discard pile, announces that they are exchanging it, and then draws one replacement card before proceeding with their turn as usual.

The game includes various booster packs that introduce additional strategic elements, increasing engagement and complexity during play.
